Fyfe has been missing since undergoing shoulder surgery in May.
Senior coach Ross Lyon said the star youngster was an automatic inclusion into the team once he was right.
“He’s certainly fit and raring to go,” Lyon said.
“We’re excited to have him back and he’ll give us an emotional lift.”
With Fyfe’s return, Cam Sutcliffe’s emergence and the likes of Neale, Nick Lower and Jesse Crichton pushing hard to play on a weekly basis, Lyon now has a plethora of midfield options at his disposal.
He said, given the emergence of certain players as viable midfield options and the weekly improvement of other stars, it had become a matter of weight of numbers.
“David Mundy is working into really good form, Stephen Hill has worked back quite well from injury and in their absence, Matt de Boer, Clancee Pearce and Tendai Mzungu have really grown and Mick Barlow is coming to hand," Lyon said.
Mayne will slot back alongside Matthew Pavlich in attack, where the pair have established a strong rapport this season.
Pavlich has kicked 45 goals, including 31 in the past six weeks, while Mayne has been deadly accurate, booting 29.3.
Freo’s leading goalkickers in 2011 were Mayne and Kepler Bradley, who each managed 25 for the season.
Fremantle defeated Port by 40 points earlier this season when the sides clashed at Patersons Stadium.
